Clinical Summary
5-FU is a chemotherapeutic agent used in the treatment of several solid tumor cancers. It is the mainstay of treatment for colorectal cancer in combination with other drugs, but is also administered for treatment of stomach, head and nec
k, breast, and pancreatic cancer. Side effects of 5-FU treatment include severe diarrhea and mucositis, as well as neurological and hematological toxicities. Although dose adjustment of 5-FU based on blood levels is not routine, clinical data demonstrate that interpatient variability can be greater than 10-fold when patients are dosed by the current standard of care. Studies have also demonstrated that 85% of colorectal cancer patients do not receive the optimal dose of 5-FU and that blood-level-guided dosing can result in less severe side effects, improved overall response, and increased survival.
